For the "full" Knightfall ending, yes you need to stop the riddler. For the "normal" Knightfall ending, you only need to complete 10 missions. Expansion missions do not count to the Knightfall, only for the 240% Batsuit

Once you have finished the main storyline and completed at least seven of the ‘Most Wanted’ missions, you are able to activate the ‘Knightfall Protocol’ from the mission select screen. This will start the epilogue.
